Access through your institution Buy or subscribe Now, Søren Knudsen from Carlsberg Research Laboratory, Denmark, and colleagues have developed a high-throughput approach to efficiently
screen large variant populations for targeted identification of single-nucleotide mutations. This tool, designated FIND-IT (Fast Identification of Nucleotide variants by droplet DigITal
PCR), uses high-throughput droplet digital PCR that enables the screening of DNA pools with a high number of individuals. Combined with the pool-and-split sampling strategy, FIND-IT can
narrow down the large genome pool of more than 500,000 individuals to a single pool and then identify the variant of interest in less than 10 days. Utilizing barley as an example, the team
identified more than 125 targeted gene knockout and missense variants, as well as miRNA and promoter gain-of-function mutations, covering genes of crop performance, grain morphology and
grain quality. Knudsen and the team also demonstrated the potential of the application on wheat, yeast (_Saccharomyces cerevisiae_) and bacteria (_Lactobacillus pasteurii_). Efficiently
identifying and incorporating genetic diversity from various plant genetic sources is crucial for developing cultivars. The FIND-IT tool improves the effectiveness and precision of
single-nucleotide mutant identification and has the potential for rapid incorporation of these variants into the breeding pipeline. This method, as well as other technologies such as
