Access through your institution Buy or subscribe Dysfunction of the synovial lymphatic system (SLS) has been implicated in the pathogenesis of inflammatory arthritis, including rheumatoid


arthritis and post-traumatic osteoarthritis (OA), but its role in age-related OA has largely remained unexplored. Evidence now suggests that impaired SLS function contributes to the


progression of age-related OA and that it could be therapeutically targeted by administration of vascular endothelial growth factor C (VEGF-C), the main growth factor for lymphatic


endothelial cells. Using a new in vivo imaging approach to measure lymphatic influx and clearance of intra-articularly administered dextran macromolecules, Lin et al. demonstrated that SLS


function is impaired in the knees of aged mice (19–24 months old) as compared with those of young mice (3 months old), with aged knees having decreased joint clearance, synovial influx and


draining lymph node capacity. The expression of _Vegfc_ and other genes related to signalling via the VEGF-C receptor VEGFR was also decreased in synovial tissue from aged mice, as
