Access through your institution Buy or subscribe Pathways related to lipid metabolism are dysregulated in cardiomyocytes in women with peripartum cardiomyopathy (PPCM), according to a new
study. Cardiomyocytes obtained from induced pluripotent stem cells (iPSC-CMs) derived from two patients with PPCM and from two healthy individuals were subjected to simulated
pregnancy-associated wall stress in vitro. RNA sequencing and gene ontology analyses indicated that pathways related to lipid metabolism were aberrantly regulated in static and stretched
PPCM iPSC-CMs compared with healthy static iPSC-CMs. Healthy iPSC-CMs had adequate metabolic plasticity via activation of glucose utilization in response to inhibition of lipid metabolism,
but this metabolic switch was impaired in PPCM iPSC-CMs. The findings were validated in an established mouse model of PPCM. Taken together, this study suggests that metabolic regulation is
