Valouev and colleagues explored how sequence-driven nucleosome-positioning preferences relate to genome function in humans by generating genome-scale nucleosome maps in three primary cell


types and by assembling of histones onto genomic DNA in vitro. From an analysis of the positions occupied by nucleosomes in vitro and in vivo, these authors describe 'container sites' in the


DNA sequence that consist of a G/C core that attracts the nucleosome flanked by A/T sequences that repel the nucleosome. In vivo, they saw phased nucleosome positioning surrounding these


sites, which supports a model — known as 'statistical positioning' — in which nucleosomes can be positioned by packing against a barrier (in this case, a sequence-positioned nucleosome at


the container site).
