ABSTRACT CULTURES of _Clostridium botulinum_ Type _D_ were grown in intussuscepted ‘Cellophane’ tubes1–3. Ammonium sulphate to 40 per cent saturation was added to the filtered dialysed

culture and the _p_H. adjusted to 5.8. The slight precipitate which formed immediately after the solution of the ammonium sulphate was found to contain most of the toxin, and was

re-dissolved and precipitated at between 25 and 30 per cent saturation at _p_H 5.8. Subsequent precipitations were carried out at between 25 and 30 per cent saturation with ammonium

sulphate, depending on the results of toxicity tests and nitrogen determinations. Purification was continued until the toxin was electrophoretically homogeneous. Diffusion measurements,

however, showed that the material was polydisperse, and that the major part diffused at a rate consistent with an average molecular weight of about a million. Access through your institution
